HIstology and cytology are integral components of cell biology. a fundamental understanding of  how to carry out a histological or cytochemical experiment is necessary to appreciate the relative strengths and weakness  of these techniques. Imagine that you are working as a researcher in a cell biology lab in which yor are interested in mapping the asymetric distribution of the plasma membrane Calcium ATPas(PMCA) and sodium, calcium exchanger(NCX) to different regions of the oplasma membrane of neurons from brain tissue slices.your advisor ask  U to design a microscopy based experiment in which you must simultaneously show the localization of these enzymes. Throughly design an experiment explaining the techniques, reagents, necessary coontrols, quantitation and statistical treatments you would need to carry out this experiment.


Any idea??????

Do some googling, pubmed literatures

start looking for immunofluorescence imaging techniques like antibody, epitope tagging, GFP fusion assays, colocalization expts

is that qualifying exam question?
the most common technique to show localization is to use immunofluorescence microscopy. you need antibodies to those enzymes. another way of addressing the same question is to construct a GFP fusion protein thet will also show locolidation. these tecq is good and reliable but not always precise. for more accurate you could do immunogold microscopy with different gold particle size.
i hope thats what you were trying to understand

no idea man!!! i give up.....but i know where vagina is ! and also know that penis and vagina wrestling creats a slippery substance which plays a vital part in formation of eggs in ovary which can indeed become a monsterous lump of living chromosome which in nine month can cry ......and shitt....
